何も指定してないのでRGBA32になってそう

Path.Combine(UnityEditor.EditorApplication.applicationContentsPath, "Tools", "PVRTexTool") で取れるPVRTexTool使うのがよいかも

ASTCについてはPVRTexToolが使えなさそうでした (astcencがないと怒られる)

Unityを調査したところunityはlibispc_texcomp.dylib (たぶん https://github.com/GameTechDev/ISPCTextureCompressor) を使っていそう。
なのでastcencをバンドルするかこれをC#から呼ぶかの二択

パディングについては調査中

左上寄せだったので端の色のコピーでpaddingすることにした。
